Let’s start with a couple of definitions.

Merriam-Webster’s words with the most vowels are words where the vowel characters are A, E, I, O, or U.

This list doesn’t include the Y or W vowels because the Y is usually a vowel combination in words with ‘FY’, ‘FYE’, or ‘Y’ sounds, and the W is often silent and doesn’t count toward the number of vowels.

The number of syllables doesn’t matter, so one-syllable words like ‘a’, ‘e’, ‘i’, ‘o’, and ‘u’ are also included.

Words with 3 vowels

The letter Y doesn’t count as a vowel, but the two vowels in the middle of the word ‘aiy’ are enough to make this word a winner with three vowels.

The same goes for ‘ea’ and ‘oi’, which are often part of words with other vowel characters.

One word with three vowels and no Y is ‘noy’, a negative word meaning ‘no’.

The word ‘oo’ can also be used to describe a cry of disappointment or frustration, but it’s usually part of the word ‘doo’, as in ‘build’ or ‘snooze’.
